10g of soil sample was collected at Cypress Nursery on 08/30/2021
-Week 0: tube contains a dense concentration of dead bacterial cells and soil debris at the bottom of the tube, having high turbidity. The tube has brown coloration.
-Week 1:The turbidity of week 1 tubes is less than week 0 tube. The reason for still having high turbidity in Week 1-tubes is there are still had other microorganisms being grown in the media tubes from the Week 0- tube. Three of four tubes have yellow coloration that means methyl parathion positive tubes and paraoxon positive tubes. That also means four tubes contain OP compound, which is being degraded into p-nitrophenol, yellow products of OP Hydrolase reaction. Tube (CSM+Px) show more yellow coloration.
-Week 2: Both paraoxon tubes show the yellow coloration, but are less intense than week 1.But the (SSM+ px) tube shows very light yellow compared to the (CSM + px) tube.Both MP tubes do not show yellow coloration.
-Week 3: there is a notice of a small change of the first tube (SSM + px) that shows only turbid white (no more yellow color). Both tubes of MP show no activity. (CSM+Px) tube shows the same result as week 2.
-Week 4:a set of week 4 tubes show the same result as week 3 tubes. But the third tube (CSM + Px) show more yellow coloration (revitalized yellow) and is clear (without turbidity). The MP tubes continue showing no activity.
-Week 5:The third tube (CSM + Px) has very light-yellow coloration as same as the first Week 2 tube (SSM + Px), and the second tube (SSM + MP) show a very slightly yellow. Both MP tubes show no activity
– After growing the sample on the GMM+ Px plate in 14 days, no OP hydrolysis occurred due to the lack of blurry-edged colonies. Colonies are only tolerant in the presence of Paraoxon.
